My Research activity to play a vital role in the field of biology through developing flexible, high performing biological technologies that Studies and Research in plant biology, plant biotechnology and genetic transformation. In a high profile international competition flooded with thousands of participants from several countries, India has once again proved it’s leadership in the area. Dr. J. Jerome Jeyakumar won the meritorious competition and established the international leadership in Biotechnology. International Agency for Standards and Ratings expressed it’s views that Dr. J. Jerome Jeyakumar efforts will explore tremendous opportunities in this segment leading it to new height. It is surely a matter of pride for civilians of India to have such brain storming intellectuals for close interaction. India has once again confirmed a rich tradition to produce world renowned intellectuals to benefit international society. In vitro Culture and Agrobacterium-Mediated Transformation Studies in Gherkin (Cucumis anguria L.) has published on International Journal of Biotechnology. International Journal of Biotechnology is World's Most Read Journal in the area of Biotechnology: World Report, 2015. Dr. J. Jerome Jeyakumar (M.Sc, B.Ed, M.Phil, M.Ed, PGDCA, DCTT, Ph.D) is Research Scholar at PG and Research Department of Botany, Jamal Mohamed College (Autonomous) Thiruchirappalli, Tamilnadu, India. He has been conferred with Fellow, IASR. My Research activity to play a vital role in the field of biology through developing flexible, high performing biological technologies that Studies and Research in plant biology, plant biotechnology and genetic transformation. Dr.J. Jerome Jeyakumar from India made a historical victory over the international competition and won Barbara McClintock Research Award-2015 in Biotechnology. Their research was highlighted in international media and gathered a large worldwide attention in scientific circles while it appeared in world's most read journal 'International Journal of Biotechnology’. In their report on In vitro Culture and Agrobacterium-Mediated Transformation Studies in Gherkin (Cucumis anguria L.) they mention that In the present study, Cucumis anguria L is a medicinally and economically important plant and also used as vegetable. We established an Agrobacterium tumefaciens- mediated transformation procedure for Cucumis anguria. Leaf explants were incubated with A. tumefaciens strain LBA4404 containing a binary vector pBAL2 carrying the reporter gene β-glucuronidase intron (GUS-INT) and the marker gene neomycin phosphotransferase (NPT-II). Following co-cultivation, leaf explants were cultured on MS medium supplemented with 6.6 µM 2,4-D combined with 3.3µM BAP containing 100 mg/l kanamycin and 300 mg/l carbenicillin. Kanamycin-resistant calluses were induced from the leaf explants after 3 weeks. Shoot regeneration was achieved after transferring the calluses onto fresh selection medium 8.8 µM BAP and 2.2 µM 2,4-D. Transgenic shoots were excised from callus and elongated in MS medium fortified with 3.0 µM GA3,100 mg/l kanamycin and 300 mg/1 carbenicillin. Finally, the shoots were rooted on MS basal medium supplemented with 3.0 µM IBA and 100 mg/l kanamycin. High transformation frequency was achieved by using 3-day-old precultured leaf explants. Further, the presence of acetosyringone (200 µM), infection of explants for 30 min and 3 days of cocultivation proved to be critical factors for greatly improving the transformation efficiency. Incorporation and expression of the transgenes were confirmed by PCR, Southern blot analysis and GUS histochemical assay. Under their leadership the research area is surely likely to expand beyond the horizons. It will be worthy to explore the further potential under their legacy. Dr. J. Jerome Jeyakumar as valuable intellectuals for the India offer exclusive opportunities for scholars around the world. One of the most attractive weightage for India is to have the awardees for promotion of the concerned scientific discipline for the benefit of its civilians. Thus India which has excellent track record to produce brain storming intellectuals, has offered a new extension in terms of Dr. J. Jerome Jeyakumar to world. The present investigation in Cucumis anguria belonging to the family Cucurbitaceae, it’s vegetable and medicinally importance. The techniques used for micropropagation, organogenesis, somatic embryogenesis, Agrobacterium mediated genetic transformation and biochemical analysis of callus. Micropropagation using the growth hormones BAP and KIN of node and shoot tip explants. Organogenesis using the growth regulators NAA, BAP and TDZ of petiole explants. Somatic embryogenesis using 2,4-D of leaf explants. Callus induced in vitro and biochemical analysis of protein, starch, amino acids and phenols of callus at 15 – 30 days. Agrobacterium mediated genetic transformation using leaf explants were confirmed by molecular analysis of GUS, PCR and Southernbloting.
